In Vitro Production System of Somatic Embryos in oenanthe Stolonifera DC .


Abstract
This study was conducted to observe the effects of medium composition and cultural environment on the uniformity and maturity of somatic embryos of Oenanthe stolonifera DC. in order to develop the rapid mass production system via somatic embryogenesis. Regenerated plantlets had a highest potential for embryogenic callus induction and 1.0 §·/§¤ 2,4-D treatment was most effective for the induction. MS liquid medium added 0.5 §·/§¤ 2,4-D was effective for the maintenance and proliferation of embryogenic callus during suspension culture. The induction and proliferation of the callus occurred to the same extent under continuous dark or 16 hr lighting condition. Formation of somatic embryo was strongly induced in a hormone-free liquid medium. Continuous subculture with the fresh medium 5 days intervals further promoted the induction of somatic embryos. Treatment of 0.05 §·/§¤ ABA at globular stage of somatic embryos was effective for increasing uniformity and inhibiting abnormality. Modified MS medium with 6% sucrose enhanced maturity of somatic embryos, whereas growth regulators had no positive effect.
